What crizotinib is?
Crizotinib is a type of drug called a tyrosine kinase inhibitor (TKI). Tyrosine kinases are enzymes (proteins that act as chemical messengers). There are many different tyrosine kinases and they can stimulate cancer cells to grow.
Crizotinib is a treatment for some people with advanced lung cancer. It is also known by its brand name Xalkori (pronounced Zal-cor-ee).
Crizotinib blocks an enzyme called anaplastic lymphoma kinase (ALK). Some lung cancer cells have an overactive version of ALK. Blocking ALK with crizotinib can stop the cells growing but it only works in cancers with the overactive enzyme. About 1 in 20 people (5%) with non small cell lung cancer (NSCLC) have the overactive ALK enzyme.
What this drug is used for:
For the treatment of non-small cell lung cancer that is caused by a defect in a gene called ALK (anaplastic lymphoma kinase). The cancer must be (ALK)-positive as indicated by an FDA-approved test.
Note: If a drug has been approved for one use, physicians may elect to use this same drug for other problems if they believe it may be helpful.
How does this drug work?
Crizotinib is a type of targeted therapy known as a kinase inhibitor. Kinases are proteins on or near the surface of a cell that transmit important signals to the cell's control center. When the ALK gene is mutated, it makes a kinase that tells the cell to grow and divide in an out of control way. Crizotinib blocks the mutated form of the ALK kinase, as well as some other kinases.
Only a small portion of non-small cell lung cancers have the mutated ALK gene. Your doctor will test your cancer cells to be sure they have this mutation before starting treatment with this drug.

Precautions:

Side effects
Get emergency medical help if you have any of these signs of an allergic reaction to Xalkori: hives; difficult breathing; swelling of your face, lips, tongue, or throat.
Call your doctor at once if you have:
headache with chest pain and severe dizziness, fainting, fast or pounding heartbeats;
very slow heart rate;
a light-headed feeling, like you might pass out;
sudden chest pain or discomfort, wheezing, dry cough or cough with mucus, feeling short of breath;
fever, swollen gums, painful mouth sores, pain when swallowing, cold or flu symptoms;
easy bruising or bleeding (nosebleeds, bleeding gums); or
liver problems--nausea, upper stomach pain, itching, tired feeling, loss of appetite, dark urine, clay-colored stools, jaundice (yellowing of the skin or eyes).
